I would like to request you if you can please include a discussion on the real life hardles we might face in setting up a cloning experiment in lab or how can we standardize a cloning protocol. Dear Saswata, Thank you for your interest. Optimizing a cloning experiment in real life lab is a challenge indeed and is difficult to discuss in a theory class. This is because every lab have their own standardized protocols which again needs to be optimized for each reaction. If you are starting it for the first time in your lab, I would suggest to follow Sambrook and Russel. Hope this helps. Best, Ma'am and Sir
